Villainous bully Karen Griffin
Karen Griffin (Kate McKinnon) is the titular villainess of "Peter's Sister", episode 14.06 of Family Guy (airdate November 15, 2015). She was the sister of protagonist Peter Griffin and, as revealed by Peter himself, was a cruel bully towards him throughout their childhood. She was also a professional wrestler who fought under the alias, "Heavy Flo".
During the episode, Karen reappeared when she came to Peter's house to spend Thanksgiving with him and his family. When Karen arrived, Karen immediately showed her bullying nature regarding her brother by passing gas in Peter's face as a "greeting". But despite Karen's behavior, the rest of the characters in attendance for Thanksgiving dinner were charmed by Karen's abrasive and witty personality.
But later, during a game of football, the evil Karen viciously tackled Peter and performed her infamous wrestling move, "The Toxic Shock", on him, wrapping her legs around his neck until he passed out. When Peter came to, he began to stutter at his villainous sister and wet his pants. This resulted in everyone (particularly Peter's daughter Meg) to finally see Karen for the callous bully she was, and Meg later encouraged Peter to finally confront his sister on how she treated him But instead, Peter decided to adopt a female persona to fight Karen in a wrestling match as a way of finally standing up to her, despite the objections from Meg and his wife Lois.
Peter (disguised under the alias "Maxi Paddy") challenged Karen just after she won a championship bet, and Karen agreed. During the fight, Karen recognized Peter, but continued the fight after Peter called her out on bullying him all their lives. Just as Karen was about to choke Peter unconscious again, Meg appeared in the ring in her own wrestling disguise "Teen LaQueefa", knocking Karen out with a chair and pinning her to the mat, winning the match. It was later revealed that Karen had been left in a coma after the fight, and the final scene of the episode had Peter refusing to get his blood type checked to help her.

- Kate McKinnon also provided the voice of the evil Régine Le Haut from the 2017 animated film Leap! and appeared on Saturday Night Live as an evil Sorceress.
